Congenital cleft of upper lip and palatine is one of the most widespread diseases of development. Children with these pathologies suffer with the respiratory and feeding functions, articulation, severe pathological changes of upper respiratory tract, therefore early surgical intervention and optimal anesthesia is important. In order to avoid postoperative complications, specific presurgical preparation - effective premedication is needed. Inhalational anesthetic halothane is widely used in pediatric surgery. According to the nowadays works, common anesthetics, including inhalational has psycho-emotional impact. Besides the regional suppressive effect, anesthesia influences on functional junction between neurons. This problem is very important because children and the elderly are most susceptible to the mentioned risks. Therefore, presurgical medication with Benzodiazepine group of drugs is used for preventing postoperative complications. It is shown that after the usage of halothane, severe mental changes, the lost of memory, including spatial memory is mentioned; these changes can be extended for more than a week after the anesthesia. As is known, the hippocampus is a part of the limbic system of the brain and is involved in emotional reactions and memory formation. According to data of last few years, GABA receptors take part in memory formation. It is likely that impact of halothane on memory should be carried out by its effect on hippocampus The Goal of the present work was to study changes of GAD65/67 and GABA positive cells quantity in rat hyppocampus in condition of halothane anesthezia with midazolam premedication. Experiments were carried out on adult and growing (20 days) rats. Study material - the brain tissue. Immunohistochemical staining method was used to assess changes in number of GAD65/67nand GABA positive cells. For estimating the proliferative activity colchicin mitotic index Immunohistochemical staining against ki67 were used.The obtained data were processed using the standard variation statistics. Data reliability was of 95-99%. Based on experimental data we have shown that halothane anesthesia induces the changes of quantity of GAD65/67 and GABA positive cells in adult rat hyppocampus. It is also determined that prevention of halothane induced changes in GABA positive cells quantity is achieved with midazolam premedication. The halothane does not cause the changes of proliferative activity of progenitor cells in dental gyrus of growing rats.
